Differentiation of pathogenic and saprophytic letospires. I. Growth at low temperatures.

The minimal growth temperature of the pathogenic leptospires is between 13 and 15 C. The saprophytic leptospires have a minimal growth temperature between 5 and 10 C, or approximately 5 C below that of the pathogens. The capability of the saprophytic leptospires to grow at temperatures below those which allow the growth of the pathogenic leptospires provides a simple method of discrimination. W...

Molecular detection of pathogenic leptospiral serovars by PCR, based on lipL21 gene

Leptospirosis is a zoonotic disease with global distribution that caused by pathogenic spirochetes of the genus Leptospira. Accurate diagnosis for differentiation of leptospirosis from other pyrogenic infections prevailing in the same locality and is imperative for proper treatment. Therefore a molecular diagnostic test with high specificity and sensitivity such as PCR is essential. Gene encodi...

Differentiation of Pathogenic and Saprophytic Leptospires with 8-azaguanine.

Johnson, Russell C. (University of Minnesota, Minneapolis), and Palmer Rogers. Differentiation of pathogenic and saprophytic leptospires with 8-azaguanine. J. Bacteriol. 88:1618-1623. 1964.-The use of the purine analogue, 8-azaguanine, as a differential agent for the separation of pathogenic and saprophytic leptospires was investigated. Growth of strains of the saprophyte Leptospira biflexa was...

Protein-free and low-protein media for the cultivation of Leptospira.

A protein-free medium composed of charcoal-detoxified Tweens (polysorbates), vitamins B12 and B1, inorganic salts, and organic buffer is described that supports the growth and subculture of pathogenic and saprophytic Leptospira. Growth was initiated from small inocula, and cell densities of 10(9) organisms per ml were attained. Antigenicity and immunogenicity of Leptospira cultivated in this me...

Rapid method for the differentiation of parasitic and saprophytic leptospirae.

The differentiation of parasitic and saprophytic leptospirae may be of importance not only from a taxonomical point of view, but it can present a diagnostic problem in culturing strains from different materials, even from human specimens (Fuzi and Cs6ka, Zentr. Bakteriol. Parasitenk., Abt. I, Orig., 178, 222, 1960). The determination of pathogenicity for laboratory animals cannot be regarded as...
